Quantification of Microstructures in Mice Alveolar Bone using Micro-computed tomography (¥ìCT)

Abstract
Periodontal inflammation increases the risk of tooth loss,particularly in cases where there is an associated loss ofalveolar bone and periodontal ligament (PDL). Histologicaland morphometric evaluation of periodontal inflammation isdifficult. Especially, the lengths of the periodontal ligamentand interdental alveolar bone space have not been quantified.A quantitative imaging procedure applicable to an animalmodel would be an important clinical study. The purpose ofthis study was to quantify the loss of alveolar bone and periodontalligament by evaluation with micro-computed tomography(micro-CT). Another purpose was to investigate differencesin infections with systemic E. coli LPS and TNF-¥áon E. coli lipopolysaccharide (LPS) in loss of alveolar boneand periodontal ligament model on mice. This study showedthat linear measurements of alveolar bone loss were representedwith an increasing trend of the periodontal ligamentlength and interdental alveolar process space. The effects ofsystemic E. coli LPS and TNF-¥á on an E. coli LPS-inducedperiodontitis mice model were investigated in this research.Loss of periodontal ligament and alveolar bone were evaluatedby micro-computed tomography (micro-CT) and calculatedby the two- and three dimensional microstructuremorphometric parameters. Also, there was a significantlyincreasing trend of the interdental alveolar process space inE. coli LPS and TNF-¥á on E. coli LPS compared to PBS. AndE. coli LPS and TNF-¥á on E. coli LPS had a slightly increasingtrend of the periodontal ligament length. The increasingtrend of TNF-¥á on the LPS-induced mice model in thisexperiment supports the previous studies on the contributionof periodontal diseases in the pathogenesis of systemicdiseases. Also, our findings offer a unique model for the studyof the role of LPS-induced TNF-¥á in systemic and chroniclocal inflammatory processes and inflammatory diseases. Inthis study, we performed rapidly quantification of the periodontalinflammatory processes and periodontal bone lossusing micro-computed tomography (micro-CT) in mice.
